چکیده

Normal human granulocytes are able to reduce the yellow dye, nitroblue tetrazolium (NBT), to insoluble blue formazan. This has been a useful reaction for the recognition of chronic granulomatous disease of childhood (CGD) and the carrier state, because granulocytes froin these patients are deficient in their ability to reduce this dye(l). It has also been suggested that granulocytes from patients with bacterial infections(2,3) and newborn infants(4,5) have increased capacity to reduce NBT, while cells from patients taking corticosteroid medications have decreased ability for NBT reduction(6). These observations are of interest because of the possible correlation of this intracellular biochemical reaction with the bactericidal capacity of the phagocyte. The interpretation of such studies is hampered by the variation in results that are obtained with different methods. It has been shown that normal, resting granulocytes in suspension reduce only small amounts of NBT, and that phagocytosis of particles seems to be necessary for NBT to gain access to the living cell(7,8). With phagocytosis of latex particles, there is a significant increase in extractable formazan from the phagocytizing cells when compared to resting

Reaction of the human granulocyte system in successfully treated bacterial bronchopneumonia.

The behaviour of peripheral blood granulocyte pools, nitroblue tetrazolium reduction by granulocytes, migration of granulocytes out of skin windows, serum muramidase activity and bone marrow reserve of granulocytes during uncomplicated bronchopneumonia were studied. The time course of changes in these parameters during infection was established, and suggests maximal changes on day 2 of infectio...

Cryopreservation of Human Granulocytes : Study of Granulocyte Function

Human granulocytes can be cryopreserved with dimethyl sulfoxide (DMSO) at 80’C. However, the percent recovery of functional cells has been unsatisfactory to date. Throughout this study we have been able to prepare relatively pure granulocytes ( 85%). cryopreserve them with 5%-1O% DMSO with and without serum, and store them at -80’C for up to 4 mo. The parameters studied were absolute cell count...

Cryopreservation of human granulocytes: study of granulocyte function and ultrastructure.

Metabolic activity of phagocytosing granulocytes in chronic granulocytic leukemia: ultrastructural observation of a degranulation defect.

The functional capacities of granulocytes in patients with chronic granulocytic leukemia are still a subject of controversy, probably due to the heterogeneity of the abnormalities observed from patient to patient. For a better definition of these abnormalities, 14 patients with untreated chronic granulocytic leukemia were studied. The patients were divided into three groups on the basis of the ...

Effect of phagocytosis on the reduced soluble sulfhydryl content of human granulocytes.

The role of reduced glutathione in relation to hexose monophosphate shunt activity and peroxide detoxification has been well established in human erythrocytes. Less is known about the content of reduced glutathione in phagocytic leukocytes and the changes that occur during functional activity. We have measured the reduced sulfhydryl content of normal resting human granulocytes and of cells isol...
